NAD+ / MOTS-c / 5-Amino-1MQ dosage by administration route
NAD+ · Injectable
Reported-use amounts for NAD+ are 50 mg per administration, 3x weekly, for 30 days. An additional reported subcutaneous range is 50–100 mg daily, without a fixed course length. This differs from intermittent injection schedules and should not be combined with an IV infusion amount.[26][29]
Direct NAD+ is the coenzyme itself, rather than the precursors NMN or NR. IV, subcutaneous, nasal and skin preparations have different concentrations and exposure patterns.[1][2][10][22][23][26]
NAD+ · Nasal
Reported-use amounts for NAD+ are 30 mg per administration, daily, on an ongoing basis.[26]
Direct NAD+ is the coenzyme itself, rather than the precursors NMN or NR. IV, subcutaneous, nasal and skin preparations have different concentrations and exposure patterns.[24][25][10][1][2][26]
NAD+ · Topical
Reported topical NAD+ routines use 100 mg per application once daily on an ongoing basis.[26]
Direct NAD+ is the coenzyme itself, rather than the precursors NMN or NR. IV, subcutaneous, nasal and skin preparations have different concentrations and exposure patterns.[24][25][1][2][10][26]
NAD+ · Intravenous
Reported-use amounts for NAD+ are 750 mg per administration, as a single infusion, during a single visit. An alternative schedule uses 250–500 mg per IV infusion over 1–2 hours, one or two times weekly, without a fixed course length. This is a reported infusion pattern, separate from the documented 750 mg single-infusion exposure and from subcutaneous injection amounts.[26][29]
An NAD+ infusion requires its own dilution, infusion duration and clinical monitoring. An injectable reminder amount does not specify an IV infusion procedure.[1][2][10][26]
MOTS-c · Injectable
Reported injectable protocols use 5–10 mg per administration, daily for 8-12 weeks. A lower-frequency reported subcutaneous pattern uses 2.5 mg per injection two times weekly; the same source also lists a two-to-three-times-weekly frequency over 8–12 weeks. This should be read separately from a daily schedule rather than combined with its daily amount.[3][16][17][28]
MOTS-c is a mitochondria-derived peptide involved in metabolic stress and AMPK-related signaling. Its proposed purpose concerns insulin sensitivity and muscle-energy adaptation.[3][16][4][17]
5-Amino-1MQ · Oral
Reported-use amounts for 5-Amino-1MQ are 50–100 mg per administration, daily, for 8–12 wk.[26]
5-Amino-1MQ inhibits nicotinamide N-methyltransferase and affects methylation and NAD-related metabolism. It is a small molecule rather than a peptide; a human fat-loss benefit has not been established.[8][20][7][21][26]
Frequency and timing
NAD+: Morning is a listed option for subcutaneous, nasal and topical use. The listed single-infusion IV exposure spans 6 hours; other IV patterns follow their own infusion durations.[24][25][26]
MOTS-c: Common timing is morning or before a workout.[17]
5-Amino-1MQ: Morning is the listed timing for the oral pattern.[9][26]
Cycle length and breaks
NAD+: Injectable: 30 days. Nasal: Ongoing. Topical: Ongoing. Intravenous: Single visit. Ongoing treatment does not follow a routine on/off cycle.[26]
MOTS-c: Injectable: 8-12 weeks. A standard break interval between courses is not established.[3][16][17]
5-Amino-1MQ: Oral: 8–12 wk. A standard break interval between courses is not established.[26]

Calculate each separately prepared component using its own vial strength and final volume. A fixed-ratio blend requires the amount of every component; the total blend weight alone is insufficient.

NAD+ / MOTS-c / 5-Amino-1MQ safety and side effects
Combination and product quality
This combination has no established shared regimen. Direct NAD+ differs from NMN and NR, MOTS-c is a separate peptide, and 5-Amino-1MQ is an NNMT inhibitor. Product identity and stability, glucose-lowering medicines and aggressive calorie restriction are relevant to risk. Component amounts cannot be added to establish combination safety.[1][2][5][9]
Side effects and long-term safety
NAD+: A reliable common-adverse-effect rate, long-term safety profile, and equivalence across formulations have not been established. Direct NAD+ has limited route-matched human safety data. Product quality is a material risk for sterile injectables because FDA has documented serious reactions linked to excessive endotoxin in compounded NAD+ products.[14][15][11][12][13]
MOTS-c: Local injection-site tolerance is a practical concern. Chronic human safety, immunogenicity, long-term mitochondrial-signaling exposure and durable metabolic effects remain unresolved.[16][17][18][19]
5-Amino-1MQ: Human pharmacokinetics, active dose, accumulation, washout, dose-response and long-term exposure are not characterized. Product identity, gastrointestinal or neurologic tolerability, liver metabolism, methylation and cancer-related biology are key considerations for oral products.[21][9]
Route-specific dosing limits
Separate products and fixed-ratio blends have different preparation requirements. Keep the exact formulation, component amount and route with each recorded dose. Reported-use schedules for NAD+ (injectable, nasal, and intravenous) and 5-Amino-1MQ (oral) are not established human treatment regimens.[26][29]

Common NAD+ / MOTS-c / 5-Amino-1MQ dosage questions
Does NAD+ / MOTS-c / 5-Amino-1MQ have one combined dose?
Each component has its own amount and schedule. A stack name does not define a blend ratio or combined dose.
Can the component doses be calculated separately?
Yes, for separately prepared components. Use each vial's peptide amount and final volume in the calculator. A combined vial requires its exact component composition.
